紫杉醇对卵巢癌细胞基质金属蛋白酶-2、9基因表达的影响

摘    要:目的:研究紫杉醇对HO8910人卵巢癌细胞的体外杀伤作用;紫杉醇对MMP-2、MMP-9 mRNA表达的影响。方法:采用MTT法检测紫杉醇对HO8910细胞的抑制率;采用半定量RT-PCR法检测紫杉醇作用于HO8910细胞时MMP-2、MMP-9基因mRNA水平的变化。结果:紫杉醇在达10-7mol/L以上浓度时对HO8910细胞抑制率>30%(P<0.01);10-8mol/L以上浓度时紫杉醇可下调MMP-2、MMP-9 mRNA的表达,其作用呈剂量依赖性。结论:在10-7mol/L以上浓度,HO8910细胞对紫杉醇具有药物敏感性;紫杉醇可能通过抑制MMP-2、MMP-9 mRNA的表达,抑制肿瘤的侵袭及转移。

Abstract:Objective: To investigate the effects of paclitaxel on MMP-2,9 expression in ovarian carcinoma cells, and the possible underlying mechanism. Methods: The effect of paclitaxel on the proliferation of H08910 cell in vitro was determined by MTT(Thiazolyl blue) assay.The expressions of MMP-2,9 mRNA in H08910 cell were assayed by RT-PCR. Results: The IC50 of paclitaxel in H08910 cell were 6.22 x 10-7mol/L.The difference of growth inhibitory rate between 10-8mol/L paclitaxel and 10-7mol/L paclitaxel was significant(P<0.01) . Paclitaxel could down-regulate the expression of MMP-2,9 mRNA when the concentration was >10-8 mol/L. Conclusions: Ovarian carcinoma cell HO8910 is sensitive to paclitaxel when the concentration is >10-7 mol/L. Paclitaxel can inhibit the expression of MMP-2,9 mRNA. It may have some influence on invasion and metastasis of ovarian carcinoma.
