鼠疫诊断芯片的探针制备

摘    要:目的 探索制备鼠疫诊断芯片特异性探针的新方法。方法 直接采用限制性核酸内切酶Sau3A Ⅰ消化鼠疫耶尔森菌3个致病性质粒,克隆在T载体上,再经巢式PCR扩增制备成打印芯片的探针。结果 该法收集到250条探针用于打印芯片。分别与鼠疫耶尔森菌,同属的假结核耶尔森菌和小肠结肠炎耶尔森菌杂交,可筛选出杂交信号强阳性的特异点阵。结论 该法简便可行,适于制备鼠疫诊断芯片的探针。

Abstract:Aim To explore a novel approach to prepare the specific DNA microarray probes for identification of Yersinia pestis Methods 3 pathogenic plasmids,which contained in Yersinia pestis,were digested with restriction enzyme Sau3A I Digested DNA fragments were cloned into pMD18 T vector and amplified with nestd PCR to prepare probes for the microarray Results 250 probes were collected by this method,a number of probes with strong positive hybridization signal were found on the DNA microarray with Yersinia pseudotuberculosis and Yersinia enterocolitica were used as negative control,respectively Conclusion This method is very convenient and suitable for the preparation of DNA microarray probes for the detection of Y pestis
