黄芩苷对T淋巴细胞增殖与活化的影响

摘    要:目的:研究黄芩苷对小鼠T细胞体外活化和增殖的影响,并探讨其免疫调节机制。方法:以刀豆蛋白A(ConA)或佛波醇酯(PMA) 离子霉素(ionomycin,Ion)诱导小鼠淋巴细胞中T细胞增殖,用MTT法测定细胞增殖;利用ConA或PMA诱导T细胞激活,流式细胞术检测T细胞早期活化标志CD69分子与CD25分子表达情况。结果:在ConA或PMA Ion刺激48h后,小鼠淋巴细胞MTT吸光度显著高于对照组(P<0.01)。黄芩苷(10~40uM)显著降低了ConA或PMA Ion引起的MTT吸光度增加(P<0.01)。在ConA或PMA刺激18h后,小鼠T细胞CD69表达率显著提高(P<0.01),ConA或PMA刺激36小时后,T细胞CD25表达率显著提高(P<0.01),黄芩苷(10~40uM)能显著降低活化(P<0.01)。结论:黄芩苷可以显著抑制多克隆刺激剂ConA或佛波醇酯(PMA)介导的小鼠T细胞的活化和增殖。

Abstract:Aim:To investigate the effects of baicalin on the expression of CD69, CD25, and proliferation of T cells by polyclonal stimulas in vitro, and explore the molecular mechanism of Baicalin. Methods: Fluorescence conjugated monoclonal antibodies and flow cytometry were used to detect the express of CD69 and CD25 by activated T cells in vitro in response to oncanavalin (Con A) or ( PMA). Mouse T lymphocyte's activation and proliferation was stimulated by ConA or PMA plus Ion, MTT assay was adopted to analyze the proliferation of mouse lymphocytes. Results: under ConA or PMA plus Ion treatment , MTT light absorption of ConA group or PMA plus Ion group was significantly higher than control group( P < 0.01) . Baicalin could significantly reduce the value ( P < 0.01). Baicalin inhibited the expression of CD69 and CD25 in activated T cells in response to Con A or PMA in a concentration-dependent manner. Conclution: Baicalin can significantly inhibit ConA or PMA mediated mouse T lymphocyte's activation and proliferation.
