葎草总黄酮对溃疡性结肠炎模型大鼠保护作用的研究

摘    要:目的:研究葎草总黄酮(HTF)对三硝基苯磺酸(TNBS)诱发大鼠溃疡性结肠炎(UC)的保护作用。方法:采用TNBS灌肠制备UC大鼠模型。实验分为6组,即正常对照(等容生理盐水)、模型(等容生理盐水)、柳氮磺胺嘧啶(SASP,300mg.kg-1)和HTF高、中、低剂量(450、300、150mg.kg-1)组,ig给药,每天1次,连续3周。给药结束后观察各组大鼠的临床表现和结肠损伤的情况,测定结肠组织中超氧化物歧化酶(SOD)、髓过氧化物酶(MPO)活性和丙二醛(MDA)含量,称定胸腺、脾脏的重量。结果:与模型组比较,HTF高、中、低剂量组大鼠的临床症状显著改善,结肠黏膜的损伤程度显著减轻,病变结肠组织中MDA含量显著减少,MPO活性显著降低,SOD活性显著增强,胸腺萎缩与脾脏肿大现象明显改善,尤以HTF高剂量组效果显著。结论:HTF对TNBS诱发的UC模型具有显著保护作用,其作用机制可能与抗炎和抗自由基氧化作用有关。

Study on the Protective Effect of Total Flavones from Humulus scandens on Ulcerative Colitis Model Rats

Abstract:OBJECTIVE: To explore the protective effect of total flavones from Humulus scandens on trinitrobenzene sulphonic acid (TNBS)-induced ulcerative colitis (UC) rats. METHODS: UC rat model was established by TNBS enema. The experimental animals were divided into 6 groups: normal control group (equal volume normal saline), model group (equal volume normal saline), SASP group (300 mg-kg^-1) and HTF high-dose, medium-dose and low-dose groups (450,300,150 mg. kg^-1). Those groups were given relevant medicine intragastrically once a day for 3 weeks. Clinical manifestation and colon injury of those groups were observed at the end of the medication. The activity of superoxide dismutase (SOD), myeloperoxidase (MPO) and the content of malondiadehyde (MDA) were determined, and the thymus and spleen of rats were weighted respectively. RESULTS: Compared with model group, clinical symptoms of HTF high-dose, medium-dose and low-dose groups were improved significantly, while colon injury relieved markedly. MPO activity of colonic tissues and the content of MDA decreased significantly, meanwhile the activity of SOD increased. Atrophy of thymus gland and splenomegaly were improved significantly, especially in HTF high-dose group. CONCLUSION: HTF shows beneficial effects on the UC model induced by TNBS. The mechanism of the action may relate to anti-inflammatory effect and anti-oxidation.
