摘    要:目的 探索含Ⅰ型血小板结合蛋白基序的解聚蛋白样金属蛋白酶(ADAMTS-1)对体外小鼠成纤维细胞(NIH3T3)转化生子因子(TGF-β1)及其下游信号通路磷脂酰肌醇3-激酶/蛋白激酶B(PI3K/AKT)的影响.方法 体外培养小鼠成纤维细胞,以ADAMTS-1为处理因素,按照不同浓度,将NIH3T3分为空白组、低浓度组(1 nM)、中浓度组(10 nM)、高浓度组(20 nM),通过RT-PCR检测细胞TGF-β1mRNA的表达情况,利用Western-blot观察细胞TGF-β1及磷酸化蛋白激酶B(pAKT)的蛋白表达情况.结果 根据RT-PCR图像显示,与空白组相比,其他各组TGF-β1mRNA的表达均下降,且随着浓度的增高,呈逐渐降低趋势(P〈0.05).根据Western-blot图像显示,与对照组相比,其他各组TGF-β1、pAKT的蛋白表达均下降,且随着浓度的增高,呈逐渐降低趋势(P〈0.05).结论 ADAMTS-1可能下调TGF-β1及其下游PI3K/AKT信号通路.

Abstract:Objective To investigate the influence of ADAMTS-1 on transforming growth factor and its downstream signal pathway PI3K/AKT in fibroblast of mouse. Methods The lung fibroblasts of mouse was cultured in vitro, and stimulated by ADAMTS-1. Based on different concentrations, they were divided into four groups: the control group, the low concentration group (1nM), the medium concentration group (10nM), and the high concen- tration group (20nM). TGF-131 and pAKT were detected by RT-PCR and western blot. Results The RT-PCR image showed that the level of TGF-β1 mRNA decreased more significantly in the rest groups than in the control group (P 〈 0. 05). With the increase of the concentration, the level of TGF-β1 and pAKT decreased gradually. Conclusion ADAMTS-1 can down-regulate the expression of TGF-β1 and PI3K/akt.
