摘 要: | ①目的研究一氧化氮合酶抑制剂N-硝基-L-精氨酸甲酯对实验性大鼠隐睾生精细胞凋亡过程中B细胞淋巴瘤/白血病-2(Bcl-2)表达的影响。②方法 42天龄雄性SD大鼠建立单侧隐睾模型,随机分为隐睾组、隐睾+L-NAME组、隐睾+生理盐水(溶媒)组、假手术组,每组12只大鼠,隐睾+L-NAME组和隐睾+生理盐水组大鼠术后分别腹腔内注射L-NAME或生理盐水,每天1次,定时定量(50mg/Kg),术后第7天分别采用RT-PCR和免疫组织化学方法检测各组隐睾Bcl-2mRNA和蛋白表达的变化。③结果与假手术组睾丸相比,隐睾组、隐睾+生理盐水组睾丸生精上皮和睾丸间质细胞内Bcl-2mRNA和蛋白表达降低,差异有显著性(P〈0.05);而隐睾+L-NAME组Bcl-2 mRNA和蛋白与假手术组睾丸无明显差别。④结论 L-NAME可通过调节Bcl-2的表达抑制热应激导致的生精细胞凋亡。

Abstract: | Objective To investigate the effect of L-nitroarginine methyl ester(L-NAME) on the expression of Bcl-2 in apoptosis of spermatogenic cells in the rat cryptorchidism.Methods Unilateral cryptorchidism model was surgically induced in the 42-day old rats and randomly divided into cryptorchidism model group,cryptorchidism group+ L-NAME group,cryptorchidism+ saline group and sham operation group(each group 12 cases).Except for the sham operation group and cryptorchidism group,the rats were injected intraperitoneally normal saline and L-NAME50mg/(Kg·d) respectively.The mRNA and protein expression of Bcl-2 in the rat cryptorchidism was detected by RT-PCR and immunohistochemical SABC assays on the 7th day after the operation.Results Compared to the sham operation group,the mRNA and protein expression of Bcl-2 in spermatogenic epithelium and Leydig cells decreased significantly in the cryptorchidism group and cryptorchid + normal saline group(P〈0.05),while there was no significant difference between the cryptorchid testes treated with L-NAME and the sham operation group.Conclusion L-NAME could inhibit spermatogenic cell apoptosis by promoting Bcl-2 expression in rats experimental cryptorchidism. |
