妇科肿瘤组织中细胞凋亡相关基因表达及其与mtDNA突变相关性的研究

摘    要:目的 研究妇科肿瘤患者原发性肿瘤组织细胞凋亡相关基因bcl-2,bax,caspase3的表达及其与mtDNA突变的相关性。方法 取32例妇科恶性肿瘤组织进行研究。应用RT-PCR方法检测bcl-2,bax,caspase3基因的表达。采用PCR-SS-CP及DNA测序检测mtDNA突变。结果 在妇科恶性肿瘤组织中,bcl-2,bax,caspase3基因的表达率分别为78.1%,43.8%及34.4%,bcl-2与caspase3,bax表达之间呈明显的负相关,Caspase3与Bax基因表达的共同阳性率和共同阴性率分别为71.4%及94.4%。在mtDNA突变的22例中,20例(90.9%)同时检测到bcl-2的表达,Bax及caspase3基因在mtDNA突变组的表达率分别为27.3%和13.6%。明显低于非突变组。结论 在妇科恶性肿瘤组织中存在bcl-2基因的高表达和bax,cas-case3基因的低表达,bcl-2与aspase3,bax表达之间呈明显的负相关,caspase3与bax基因存在共表达关系。细胞凋亡相关基因表达与mtDNA突变亦存在一定的相关性。

Abstract:Objectives: To investigating the gene expression of BCL?2, BAX, and Caspase3 in tumor tissues of gynecologic oncology patients and their relationship to mtDNA mutation. Methods: Total 32 samples of gynecological malignant tumors were taken. RT?PCR were done to examine bcl?2?bax?caspase3 gene expression. PCR?SSCP and DNA sequence were done to detect mtDNA mutation. Results: The rates of bcl?2, bax, and caspase expression in gynecological malignant tumors were 78.1%(25/32),43.8%(14/32),and 34.4%(11/32)respectively. There were negative correlation among bcl?2,caspase3, and bax. The rates of co?positive and negative of caspase3 and bax were 71.4% and 94.4% respectively. In total 22 cases of mtDNA mutation, 20 cases(90.9%) had bcl?2 co?expression. Bax and caspase3 gene expression in the mtDNA mutation group were 27.3% and 13.6% respectively. They were much lower than that of mtDNA non?mutation group. Conclusions: There are high expression of bcl?2 gene and low expression of caspase3 and bax gene in gynecological malignant tumors. The expression of Bcl?2 had a negative relationship with expression of bax and caspase3. But bax and caspase3 have co?expression. There are positive relationship between cell apoptosis associated gene expression and mtDNA mutation.
