摘    要:目的探讨三氧化二砷(As2O3)及其联合地塞米松(DEX)对胶质母细胞瘤细胞的增殖抑制作用。方法实验分为空白对照组、As2O3组、DEX组、As2O3-4-DEX组,经倒置显微镜观察细胞形态、细胞计数、MTT法、流式细胞计数等方法研究As2O3及其联合DEX作用对胶质母细胞瘤细胞的增殖抑制作用。结果As2O3能够且呈浓度和时间的依赖性抑制BT-325细胞的增殖;DEX具有明显的协同效应。结论As2O3及其与DEX联合用药可能成为治疗胶质母细胞瘤的一条新途径。

Abstract:Objective To evaluate the inhibitory effects on proliferation of human glioblastoma cell line BT-325 in vitro by arsenic trioxide (A_(S2)O_3) alone and in combination with dexamethasone (DEX) on.Methods According to different agents added in the media of cultured BT-325 cells, cultured glioblastoma cells were allocated into A_(S2)O_3, DEX, A_(S2)O_3+DEX and untreated control groups. The cell growth was assessed through morphology observation by invert microscope, cell count, MTT assays and flow cytometric.Results A_(S2)O_3 effectively inhibited the proliferation of BT-325 cells in a dose- and-time-dependent manners. On the other hand, this study showed the significant synergistic inhibitory effect of A_(S2)O_3 with DEX on proliferation of BT-325 cells. Conclusion Administration of A_(S2)O_3 with DEX may become an alternative method to the treatment of glioblastoma.
