大鼠三叉神经脊束尾侧亚核内Cdk5/p35的表达与活性

摘    要:目的 检验Cdk5/p35在神经元成熟和萌芽中扮演着重要作用的假说。方法 采用Western blot实验、免疫沉降和Cdk5活性分析的方法,研究了出生后各发育阶段大鼠三叉神经脊束尾侧亚核(Vc)内该激酶的活性变化和Cdk5、p35的表达水平。结果 在大鼠三叉神经脊束尾侧亚核组织中,p35的表达随着发育阶段递增而减少,新生大鼠p35的表达最高,到正常成体大鼠几乎无p35的表达。Cdk5的表达量在各发育阶段中是恒定不变的。新生大鼠中的Cdk5活性比成体正常大鼠中的Cdk5活性约高六倍,Cdk5的活性变化与p35的表达变化具有一致性。结论 提示Cdk5/p35与三叉神经脊束尾侧亚核内神经元的分化、成熟及可塑性有密切的关系。

Abstract:Objective To test the hypothesis that Cdk5/p35 plays important roles in neuronal maturation and sprouting. Methods We studied the changes of Cdk5 activity, expression levels of Cdk5 and p35 in the development in spinal trigeminal subnucleus caudalis(Vc) by Western blot method,Immunoprecipitation and Kinase assay. Results By degrees the expression of p35 significantly decreased from newborn to Vc of adult rats. The expression of p35 was mostly prominent in Vc of newborn rats, and it was the most lowest in Vc of adult rats, In contrast, the expression of Cdk5 was constant in Vc throughout the developmentrats. Cdk5 activity in the newborn rats was about 6 times the height of that one in Vc of normal adult rats. The changes of the kinase activity coincided well with those of p35 expression in the development rat. Conclusion These data suggest Cdk5/p35 plays important roles on synaptic reorganization of Vc, expression of p35 may be a novel regulatory mechanism to control Cdk5 activity in Vc.
