PC12细胞缺糖损伤的分子生物学特征

摘    要:目的在体外模拟能量代谢障碍研究PC12细胞缺糖损伤的分子生物学特征.方法电镜观测形态改变,MTT检测功能异常,RT-PCR测定Bcl-2在RNA水平上的表达.结果电镜观察到细胞有凋亡现象;MTT显示细胞活力降低;RT-PCR结果表明Bcl-2在6~16 h表达上调.结论缺糖损伤的PC12细胞既有形态的变化也有功能上的变化,并同时伴随着凋亡和坏死两种死亡现象.

Abstract:Objective By using glucose deprivation (an insult relevant to the brain ischemia) media,PC12 cell strain was evaluated.Methods By cellular and molecular biological techniques including electron microscopy,MTT,and RT-PCR to meaure and observe.Results Electron microscopy reveals the treated PC12 cells underwent apoptosis;MTT result shows the treated PC12 cells viabilities induced;Bcl-2 RNA levels expressed to peak levels 6-16 hours after glucose deprivation treatment and then declined gradually.Conclusion The PC12 cells deprived of glucose underwent the morphological and functional alterations including apoptosis and necrosis.
