羟基喜树碱和阿霉素不同时间序贯给药对小鼠移植性肿瘤疗效的影响

摘    要:目的 探讨羟基喜树碱(hydroxycamptothecin,HCPT)与阿霉素(adriamycin,ADM)联合用药时最佳给药顺序及时间间隔。方法 小鼠腹腔内接种S180腹水,随机分为8组,腹腔给药,包括注入生理盐水组(对照组),HCPT单药组(10mg/kg),ADM单药组(5mg/kg),以及注入HCPT用药后0、8、12、24、72h后分别给ADM,观察小鼠的体质量,生存天数及治疗结束时存活小鼠的数目,计算生命延长率及治愈率。结果 单用HCPT组无效。ADM组及5个序贯给药组有一定的疗效。但HCPT与ADM同时给药组的疗效低于单用ADM组,HCPT与ADM序贯给药中以间隔8h的小鼠生命延长率和治愈率最高,其次为12h组,结论 HCPT与ADM序贯给药有协同作用,以间隔8、12h作用最强。

Abstract:ObjectiveTo investigate antitumor effect of sequential and interval administration of hydroxycamptothecin(HCPT) and adriamycin(ADM).MethodsMice implanted with murine sarcoma S 180 cell were divided randomly into eight groups, then administrated by ip of 0.9 % NaCl(control), single HCPT(10 mg/kg),single ADM (5 mg/kg) and sequential administrations of HCPT followed by ADM after 0,8,12,24,72 h, respectively. Body mass, survival days and cured numbers were observed, then life span increase(LSI) rate and curable rate were evaluated.ResultsSingle HCPT was ineffective. Single ADM and the other five groups of sequential administration were shown certain effects. But simultaneous administrations of HCPT and ADM was not superior to single ADM. Sequential administration of HCPT and ADM, especially interval of8 h, showed the highest LSI rate and curable rate,the interval of 12 h was also higher.ConclusionSequential administration of HCPT and ADM showed the synergy effects,especially,8-12 h.
