免疫负调控分子TIPE2的研究进展

摘    要:肿瘤坏死因子α诱导蛋白8-2(tumor necrosis factor-α-induced protein-8-like2,TNFAIP8L2,TIPE2),是最近发现的一种新型免疫负调控分子,优先表达在正常小鼠的髓系和淋巴系细胞,而在人体分布更加广泛。TIPE2可以调控T细胞受体(TCR)和TOLL样受体(toll-like receptor,TLR)的信号转导,从而对固有性免疫应答和适应性免疫应答进行免疫负调控,以便维持免疫自稳;TIPE2还可以通过直接结合caspase-8后,抑制激活蛋白(activator protein,Ap)-1和核因子(nuclear factor,NF)-κB的活化等途径发挥其促细胞凋亡作用。现研究已证实TIPE2的表达异常与系统性红斑狼疮(systemic lupus erythematosus,SLE)等多种疾病有关。本文总结了TIPE2基因定位结构特点、生物学功能以及TIPE2异常表达和疾病的关系和临床意义。

Abstract:Tumor necrosis factor-α-induced protein-8-like2(TNFAIP8L2),is a recently discovered novel immune negative regulatory molecule.TIPE2 can transduce signal on T cell receptor(TCR)and Toll-like receptor(TOLL)simultaneously.As a result,inherent immunity and adaptive immunity will be in negative immunoregulation thus keep the immune homeostasis.In order to promote apoptosis,TIPE2 even can depress the activation of activator protein (Ap)1 and nuclear factor(NF)-κB after TIPE combine with caspase-8 directly.Now the research have confirmed that the abnormal expression of TIPE2 relate to systemic lupus erythematosus(SLE) and the other diseases.This article elaborate the newest progression of the structure of gene TIPE2 and its biological function.Also this paper make a simple summary with the abnormal expression of TIPE2,relationship with diseases and its clinical sense.
