鲜淡菜中软骨藻酸的液相色谱紫外法研究

摘    要:[目的]根据美国FDA对软骨藻酸(domoic acid)的控制指标,建立一种简便的水产品中软骨藻酸液相色谱紫外测定方法。[方法]采用含甲醇的水溶液,选择性地提取鲜淡菜中软骨藻酸,经离心、过滤等手段进行样品处理,在最优化的条件下进行液相色谱检测。[结果]本研究方法的实际测定生物样品——鲜淡菜中软骨藻酸范围为0.12.50mg/kg,最低检测限达到0.007mg/kg,并且在现有分离条件下无明显干扰物存在;同时经与原分析方法比对,具有很好的一致性。[结论]本方法建立了生物样品鲜淡菜中软骨藻酸的液相色谱紫外测定方法,具有简便、快速的特点,能适合在一般分析实验室开展此项检测工作。

Abstract:[Objective] A liquid chromatographic method was established to easily detect domoic acid in oceanic products according to the standard of FDA in USA. [Methods] The method includes the selective extraction of domoic acid in fresh mussel, the pretreat-ment with centrifuge and filter,chromatographic analysis in the optimized conditions. [Results] The detection range is from 0.12 mg/kg to 50 mg/kg in fresh mussel without obvious interferes.The method detection limit(MDLs) is 0.007 mg/kg. It shows consistency with the original method by comparison. [Conclusion] The paper introduces an analytical method to detect domoic acid in marine products by HPLC/UV.As its simplicity and speediness,the method is suitable for an ordinary analysis in chemical lab.
