人参皂苷Rg1对过氧化氢诱导PC12细胞氧化损伤的保护作用

摘    要:观察人参皂苷Rg1(GSRg1)对H2O2诱导的PC12细胞氧化损伤时细胞存活率、DNA及蛋白的影响。GSRg1与PC12细胞共同培养24h,经H2O2 处理30min,通过显微镜观察细胞的形态,MTT法测定细胞生存率,用单细胞凝胶电泳法测定DNA单链损伤,ELISA方法测定细胞内的羰基蛋白含量。结果:经H2O2 处理后细胞生存率明显降低,细胞内DNA单链损伤及羰基蛋白含量明显升高,并随H2O2浓度的增加而加重。而经GSRg1预培养的细胞生存率明显升高,细胞内DNA单链损伤及羰基蛋白含量明显减轻,其作用呈剂量依赖关系。结论:GSRg1通过降低H2O2 对PC12细胞内DNA和蛋白的氧化损伤而起到对神经细胞的保护作用。

Abstract:To study the effects of Gensenoside Rg 1 (GSRg1) on the cell viability, DNA damage and protein carbonyl formation induced by H2O2 in PC12 cells, the cells were treated with H2O2 for 30 min after incubated with GSRg1 for 24 hours. The morphology of the cells was observed by microscope and the cell viability was measured by MTT method, DNA single breaks were determined by Comet assay and the protein carbonyl contents by ELISA. Results: The cell viability was decreased obviously, DNA damage and the protein carbonyl formation were increased and aggravated along with the enhancement of H2O2 concentration. GSRg1 increased the cell viability markedly, protected the cells from H2O2 -induced DNA damage and protein carbonyl formation in a dose-dependent manner. It is shown that GSRg1 can protect the PC12 cells against oxidative injury by reducing cellular DNA and protein damage.
