姜枣合用对D-半乳糖致衰小鼠钙稳态影响的实验研究

摘    要:目的:观察:1姜枣合用对维持细胞内钙稳态的作用2中药维持细胞内钙稳态机制的初步探讨。方法:本实验以D-半乳糖衰老小鼠为研究对象,以按照不同比例姜枣混合的水煎剂灌胃4 5 d,测定了青年、老年小鼠红细胞超氧化物歧化酶(SOD)活性,心肌线粒体内丙二醛(MDA )含量,心肌细胞膜Na - K - ATPase、Ca2 - ATPase活性,心肌组织及线粒体Ca2 含量变化。结果:姜枣合用可提高小鼠红细胞SOD活性,心肌细胞膜Na - K - ATPase、Ca2 ATPase活性、心肌线粒体Ca2 含量,并能降低心肌线粒体内MDA含量、心肌组织Ca2 含量。在各给药组间无显著性差异。结论:1姜枣合用可维持细胞内钙稳态,具有一定的抗衰老作用;2自由基与钙超载协同作用可能是导致衰老的重要原因。

Abstract:Objective: To investigate the effects and mechanism of mixture of jujube and zingiber on the intracellular calcium homeostasis. Methods:The aging mice induced by D-gal were filled with different proportions of mixture o f jujube and zingiber for 45 days. we detected the SOD activity in RBC, the MDA level in myocardial mitochondria, the activities of Na -K -ATPase and Ca 2 -ATPase in the myocardial cell membrane, the calcium concentration change i n the myocardial tissue and mitochondria, and at the same time, the effects of d ifferent proportions of mixture of jujube and zingiber on this indexes were obse rved. Results:The SOD activity in RBC, the activities of Na - K -ATPase and Ca 2 -ATPase in the myocardial cell membrane, and the calci um content in the myocardial mitochondria were decreased with ageing; meanwhile the MDA level in the myocardial mitochondria and the Ca 2 concentration in the myocardial tissue were increased with ageing. The mixture of jujube and zin giber enhanced the SOD activity in RBC, the activities of Na -K -ATPase and Ca 2 -ATPase in the myocardial cell membrane, the Ca 2 concentration in the myocardial mitochondria, and decreased the MDA level in the myocardial mi tochondria and the Ca 2 concentration in the myocardial tissue. There was no significant difference in the three group. Conclusion:1. Mix ture of jujube and zingiber can enhance the antioxidant ability, reduce the dama ge to the cell membrane of free radical(FR)and maintain the intracellular Ca 2 concentration. Therefore, it has an anti-aging effect. 2. The corporation o f FR and Ca 2 overload may be the important cause of aging.
