三七皂甙对急性脑梗塞患者血浆和脑脊液MCP-1、ET含量的影响

摘    要:研究三七皂甙对急性脑梗塞患者血浆和脑脊液单核细胞趋化因子(MCP 1)和内皮素(ET)水平在不同时段变化的影响。方法:用酶联免疫法和放射免疫法测定MCP 1和ET在不同时间血浆和脑脊液含量的变化情况,及应用三七皂甙治疗后的影响。结果:脑梗塞患者血浆MCP 1和ET的水平明显增加,三七皂甙组各时间段MCP 1和ET的含量与对照组比较有显著性差异(P<0.05),MCP 1在第3天达到峰值,第5天开始下降。ET在第3~5天达峰值,之后逐渐下降,第10天的含量仍高于第1天。脑梗塞48?h后患者脑脊液MCP 1和ET的含量亦明显升高。应用三七皂甙后血浆MCP 1和ET的水平均显著低于对照组(P<0.01)。结论: 脑梗塞后血浆和脑脊液MCP 1和ET水平明显增高(P<0.05);MCP 1和ET的变化有一定相关性(r=0.852)。三七皂甙对脑梗塞后血浆和脑脊液MCP 1和ET的表达有部分抑制作用,这可能是三七皂甙减轻脑梗塞后缺血缺氧的分子机制之一。

Abstract:Objective:To investigate the concention changes of the monocyte chemoattractant protein-1(MCP-1) and the endothelia(ET) in serum and cerebrospinal fluid(csf) of patients with acute brain infarction treated by saponins.Methods: The level of MCP-1and ET in different times in serum and cerebrospinal were determined by enzyme-linked immunoadsordent(ELLISA) and radioreceptor assay(RRA). CSF samples were obtained on the first day of the onset and at 48 hours later during the lumbar puncture procedure.Results: Both the serum levels of MCP-1 and ET in different times were notably higher in ischemic stroke than healthy man(P<0.05).The level of MCP-1 reached the peak on the day 3 and then decreased on the day 5-10,and that of ET,too.The levels of MCP-1 andET significantly increased 48 hours after the onset of brain infarction,but they significantly decreased after the treatment of saponins(P<0.05). Conclusions: Our results suggest that MCP-1 and ET may play roles in the inflammatory reaction during the early phase of ischemic stroke;the increasing of MCP-1 can stimulate the secretion and expression of ET;saponins can partly resist the secretion and expression of ET and MCP-1,which may be one of the mechanism for decreasing the danger of cerebroinfarction.
