| 摘 要： | 目的测定丽江产玛咖中必需氨基酸的含量。方法以2，4-二硝基氟苯为衍生试剂，与丽江产玛咖中18种氨基酸柱前衍生，用Waters600高效液相色谱仪，菲罗门公司Symmetry C18色谱柱，以含1％N，N-二甲基甲酰胺的0．05mol／L醋酸钠-醋酸缓冲液（pH=6．4）及乙腈：水（1：1）进行梯度洗脱，检测波长为360nm，柱温为35℃，进样量为20μL。结果18种氨基酸的线性回归方程r值为0．9995～1．0000（n=5），平均回收率为99．54％-101．84％，RSD值为0．38％～1．82％，n=6。结论本法快速、简便，结果准确。

| Abstract: | Objective To analyze the contents of 17 amino acids in MACA using pre-column derivatization method. Methods The amino acids in MACA were pretreated with 2,4-dinitro-fluorobenzene and their derivatives were analyzed on Waters 600 HPLC using Symmetry Cls column. Gradienteluents consisted of sodium acetate solution（pH=6.4） with 1% N,N-dimethylformamide, acetonitrile and water（1 ： 1）. The Detection wavelength was 360nm and column temperature was 35℃ . Results The calibration curves for 18 amino acidshad good linearity（r=0.9995 - 1,n=5）. The mean recoveries of 18 amino acids were 99.54%-101.84% with RSD 0.38%-1.82%（n=6）. Conclusion The method is quick, simple for the content analysis of the amino acids in the injections. |
