椎间盘突出症大鼠血清内皮素及降钙素基因相关肽水平变化的实验研究

摘    要:目的观察椎间盘突出症大鼠血清内皮素(ET)、降钙素基因相关肽(CGRP)的变化。方法30只雄性SD大鼠,随机分为3组,每组10只,对照组(C组)、实验3d组(T3组)和实验6d组(T6组)。T3组和T6组硬膜外腔注射20μl自体髓核混悬液和2%利多卡因30μl,C组硬膜外腔注射0.9%氯化钠溶液20μl和等量2%利多卡因。T3组观察3d,T6组和C组分别观察6d后剖胸取心室血2ml,离心后取上清液,采用放射免疫法测定血清ET、CGRP的水平。结果T3组和T6组血清ET、CGRP水平高于C组(P〈0.05或〈0.01)。结论ET、CGRP在椎间盘突出症发生发展中有重要作用。

Abstract:Objective To investigate the changes of serum levels of endothelin (ET) and calcitonin gene related peptide (CGRP) in rats with intervertebral disc herniation.Methods Thirty adult male SD rats weighed 280-320g were randomly divided into 3 groups (n=10,in each group):groupⅠ(contol group),groupⅡ(3-day experiment),group Ⅲ (6-day experiment). The rats in group Ⅰwere given 0.9% NS 20μl and 2% lidocaine 20μl injecting into epidural space at L4,5 interspace,then the rats were observed for 6 days. The rats in group Ⅱand group Ⅲ were given autologous nucleus suspension 20 μl and 2% lidocaine 30 μl injecting into epidural space,then the rats were observed for 3 and 6 days respectively.The blood samples were taken from the ventricle of heart at the end of 3 days (groupⅡ) or 6 days (groupⅠ,Ⅲ) to detect the serum concentration of ET and CGRP by radioimmunoassay. Results The serum concentrations of ET and CGRP in group Ⅱand group Ⅲ were significantly higher than those in groupⅠ(P〈0.05 or 0.01). Conclusion ET and CGRP play an important role in the pathogenesis and development of intervertebral disc herniation.
