卵巢切除大鼠脑内APP β位点裂解酶表达的变化

摘    要:目的 观察卵巢切除大鼠脑内不同部位淀粉样蛋白前体蛋白(APP)β位点裂解酶(BACE)表达的变化,探讨雌激素缺乏与APP代谢系统可能存在的内在联系。方法 成年健康雌性Wistar大鼠14只,随机分为卵巢切除组(模型组)和对照组,每组7只。BACE免疫荧光标记细胞化学染色,激光共聚焦显微镜下观察海马CA1区、皮质区BACE阳性细胞的表达情况。结果 对照组BACE阳性细胞表达极少,模型组皮质区、海马CA1区BACE阳性细胞数较对照组显著增多(t=5.59、6.27,P〈0.01)。结论 卵巢切除大鼠脑内BACE表达增多,推测雌激素与BACE存在相互作用的关系。

Abstract:Objective To investigate the possible mechanism between the loss of estrogen and amyloid precursor protein (APP) metabolizing system by observing variations of β-site APP cleaving enzyme (BACE) in different brain areas of rats after ovariectomy. Methods Fourteen Wistar rats were divided into two groups: model group (with ovariectomy), and control group (with no ovaries removed). The brain tissues were taken out and the BACE immunofluorescence staining labeled cells were observed in the cerebral cortex and CA1 section of hippoeampi under confocal laser microscopy, the number of immunofluorescence staining labeled cells of BACE between the two groups were compared. Results There were a lot of immunofluorescence staining labeled cells of BACE found in the model group than in the control group in the cerebral cortex and CA1 section of hippocampi (t= 5.59,6.27; P<0.01). Conclusion The increase of immunofluorescence staining labeled cells of BACE in cerebral cortex and CA1 section of hippocampi of rats with ovariectomy suggests that estrogen has an interaction on BACE.
