妇科肿瘤组织中端粒酶活性的研究

目的 :探讨端粒酶与妇科肿瘤发生发展的关系及其作为妇科肿瘤标志的可能性。方法 :采用端粒重复序列扩充法 ,结合银染色 ,研究 2 3例妇科恶性肿瘤、2 1例良性肿瘤及 2 2例正常组织的端粒酶活性。结果 :2 3例恶性肿瘤中端粒酶表达阳性者 2 1例 (91.3% ) ,且多数为强阳性 ,强阳性率 73.9% ,阳性表达与肿瘤发展阶段、恶性程度不相关。良性肿瘤 2 1例 ,10例弱阳性 (4 7.6 % )。正常组织 2 2例 ,5例弱阳性 (2 2 .7% )。结论 :端粒酶在妇科恶性肿瘤的发生发展中起重要作用 ,端粒酶活性的检测可能成为妇科恶性肿瘤的标志物

DETECTION OF TELOMERASE ACTIVTY IN GYNECOLOGIC TUMORS

Objective:To study the relationship between telomerase expression and development of gynecologic tumor, and to explore the diagnostic significance of the detecting of telomerase activity for gynecologic tumor.Methods:Telomerase activity in some specimens, including 23 malignant tumors and 21 benign tumors and 22 normal tissues from gynecologic diseases, were examined by TRAP-Silver assay.Results:Positive rates of telomerase activity in the three groups were 91.3%, 47.6% and 22.7%, respectively. Positive rates of telomerase activity in the malignant tumors were significantly higher than those in the and normal tissues ( P <0.001). Furthermore,most of them were strong telomerase activity, while a few benign and normal tissues were only weakly positive. There is no relationship between telomerase expression and the stage or the differentiation of malignant tumor.Conclusions:Telomerase plays an important role in the development of gynecologic tumor. Telomerase may be a useful biomarker for the diagnosis of gynecologic tumor.
