摘    要:目的:探讨黄蒲通窍胶囊对血管性痴呆大鼠学习记忆影响的机制.方法:采用线栓法栓塞大脑中动脉,复制血管性痴呆(VaD)大鼠模型,观察各组大鼠脑组织一氧化氮(NO)、一氧化氮合酶(NOS)和环磷酸鸟苷(cGMP)的变化.结果:4周后模型组大鼠脑组织NO含量、NOS活性和cGMP水平明显增高,中药组、阳性对照组大鼠NO含量、NOS活性和cGMP水平显著降低,差异均有显著性(P《0.01).结论:黄蒲通窍胶囊可减少NO路径诱导的细胞凋亡发生,降低NO的神经毒性,保护脑细胞,降低NO至适量水平,从而促进VaD大鼠学习记忆能力的改善.

Abstract:Objective:To probe into mechanisms of effects of Huangpu Tongqiao Capsule(HTC) on the contents of cyclic guanosine monophosphate(cGMP) and nitric oxide(NO) and activities of nitric oxide synthase(NOS) in neurons of rats with vascular dementia(VaD).Methods:The rat VaD model was made by embolizing middle cerebral artery with threads,and the changes of the contents of cGMP and NO and activities of NOS in neurons of VaD rats in each group were observed.Results:After 4 weeks,the contents of cGMP and NO and activities of NOS in neurons of rats increased in model group compared with those of the control group,and significantly increased in Chinese medicine group compared with those of the model group(P<0.01).Conclusions:HTC may reduce neuronal apoptosis induced by NO pathway and decrease neuronal toxicity,and enhance abilities of learning and memory by lowering the content of NO to a proper level.The abovementioned effects might be important mechanisms for the treatment of VaD.
