恶性肿瘤患者的靶向肿瘤干细胞免疫治疗疗效观察

目的探讨肿瘤干细胞（CSC）抗原负载树突细胞（DC）活化同源的细胞因子诱导的杀伤细胞（CIK）,形成CSC-DCCIK效应细胞,治疗恶性肿瘤的临床疗效。方法选取32例2011年1-12月入院治疗的均经病理检查证实诊断为恶性肿瘤的患者,其中肺癌10例,膀胱癌15例,乳腺癌7例,给予CSC-DC-CIK细胞治疗。结果共培养后CIK表面标志CD3、CD8、CD56的表达率（分别为87.74%±5.23%、82.67%±6.12%、55.62%±8.33%）高于单独培养CIK（分别为61.65%±7.79%、50.21%±4.94%、22.12%±2.85%）,差异均有统计学意义（P〈0.01）。CIK组、DC-CIK组和CSC-DC-CIK组间细胞上清中干扰素、肿瘤坏死因子α和白细胞介素2水平的差异有统计学意义（P〈0.01）。32例患者中,有效患者为3例（9.4%）,显效患者为18例（56.3%）,无效患者6例（18.7%）,5例（15.6%）患者属于其他（没有坚持治疗等）。有效和显效患者共21例,治疗有效率为65.6%（21/32）。结论靶向肿瘤干细胞免疫治疗有很好的应用前景,有待进一步研究。

Immunotherapeutic effects of targeting cancer stem cells on patients with malignant tumors

Objective To investigate the clinical thrapeutic effects of CSC-DC-CIK effector cells on patients with malignant tumor or leukemia. Methods Thirty two inpatients with malignant tumors during January to December in 2011（10 cases with lung cancer,15 cases with bladder cancer and 7 cases with breast cancer） certified by pathological examination were selected as the subjects,and all subjects were treated by CSC-DC-CIK effector cells. Results The expression rates of surface markers CD3,CD8 and CD56 in CIK co-cultured with CSC-DC were 87.74%±5.23%,82.67%±6.12% and 55.62%±8.33% respectively,which were significantly higher than those（61.65%±7.79%,50.21%±4.94% and 22.12%±2.85%） in CIK without co-culture（P〈0.01）. There were significant differences of interferon,tumor necrosis factor-α and interleukin-2 levels in cellular supernatant among CIK,DCCIK and CSC-DC-CIK groups（P〈0.01）. In 32 cases,the effective rate,markedly effective rate and invalid rate were 9.4%（3 cases）,56.3%（18 cases） and 18.7%（6 cases）,respectively. Other was 15.6%（5 cases）. The effective and markedly effective rate was 65.6%（21/32）. Conclusion Immunotherapy of targeting cancer stem cells has good prospects,but which should be studied in future.
