抗纤益心方对扩张型心肌病大鼠心肌纤维化相关蛋白表达的影响

摘    要:目的:观察抗纤益心方对扩张型心肌病(dilated cardiomyopathy, DCM)大鼠心肌纤维化的作用及可能的作用机制。方法:参照相关文献将雄性Wistar大鼠随机分为造模组和正常组,造模组大鼠用呋喃唑酮复制DCM大鼠模型,10周后根据心脏超声情况将造模成功大鼠随机分为模型组、抗纤益心方低、中、高剂量组与卡托普利组5组各10只。药物干预4周后检查大鼠心功能相关指标,Masson染色观察心肌组织病理变化,Real-time PCR法检测心肌组织心房利钠肽(atrialnatriureticpeptide, ANP)、B型利钠肽(brain natriuretic peptide, BNP)、结缔组织生长因子(connective tissue growth factor, CTGF)、α-平滑肌肌动蛋白(α-smooth muscle actin,α-SMA)mRNA的表达水平,Weston blot法检测Ⅰ型胶原蛋白(collagen Ⅰ,CoⅠ-1)、CTGF、α-SMA蛋白表达水平。结果:与模型组比较,抗纤益心方各组及卡托普利组射血分数(ejection fraction, EF)、左室短轴缩短率(fractional shortening, FS)明显升高(P<0.05,P<0.01),ANP、BNP、α-SMA、CTGF mRNA及相关蛋白表达水平都有不同程度降低(P<0.05,P<0.01),差异有统计学意义。结论:抗纤益心方能够抑制DCM模型大鼠心肌纤维化,可能与其抑制心肌成纤维细胞的增殖、转分化有关。

Abstract:Objective:To observe the effects and mechanism of Kangxian Yixin Decoction on myocardial fibrosis in dilated cardiomyopathy(DCM)rats.Methods:With reference to related literature,male wistar rats were randomly divided into model groups and normal groups.The model rats were modeled with furazolidone to replicate the DCM rat model.After 10 weeks,the model rats were randomly divided into five groups according to the cardiac ultrasound situation:the model group,the low-dose,medium-dose,and high-dose groups of Kangxian Yixin Decoction and the captopril group,with 10 rats in each group.After four weeks of drug intervention,rat cardiac function-related indicators were examined,and myocardial tissue pathological changes were observed with Masson staining.Real-time PCR was used to detect the expression levels of atrialnatriureticpeptide(ANP),brain natriuretic peptide(BNP),connective tissue growth factor(CTGF),andα-smooth muscle actin(α-SMA)mRNA in myocardial tissue.Westem blot was used to detect Collagen I(CoI-1),CTGF andα-SMA protein expression levels.Results:Compared with the model group,the ejection fraction(EF)and fractional shortening(FS)levels of Kangxian Yixin Decoction and Captopril group were significantly increased(P<0.05,P<0.01),and the expression levels of ANP,BNP,α-SMA,CTGF mRNA and related protein expression levels decreased in varying degrees(P<0.05,P<0.01).Conclusion:Kangxian Yixin Decoction can inhibit myocardial fibrosis in DCM model rats,which may be related to its inhibition of myocardial fibroblast proliferation and transdifferentiation.
