摘    要:目的:研究参麦注射液对高血压心衰大鼠粪便代谢组学的影响。方法:20只Dahl盐敏感大鼠随机分为正常组(6只)、模型组(7只)和参麦注射液组(7只),以高盐饲料(8%NaCl浓度)喂养模型组、参麦注射液组大鼠20周建立高血压心衰模型。给药15 d后收集各组粪便样本,运用基于液相色谱-质谱(liquid chromatography mass spectrometry,LC-MS)联用的代谢组学技术进行研究。结果:主成分分析、偏最小二乘判别分析得分图显示,3组大鼠的代谢模式存在显著差异,与模型组比较参麦注射液组神经氨酸、别石胆酸、丙氨酰色氨酸、3,9-十六碳二烯酸、N-棕榈酰酪氨酸、亚油基肉碱、3-羟基-顺式-5-十四碳酰肉碱表达水平回调。结论:参麦注射液干预高血压心衰的机制涉及优化氨基酸代谢、蛋白质代谢、肉碱代谢、胆汁酸代谢、脂质代谢等多个层面。

Abstract:Objective:To study the effect of Shenmai injection on fecal metabonomics in hypertensive heart failure rats.Methods:Twenty Dahl salt-sensitive rats were randomly divided into normal group(6 rats),model group(7 rats)and Shenmai injection group(7 rats).Rats in model group and Shenmai injection group were fed with high salt diet(8%NaCl concentration),and hypertensive heart failure model was established after 20 weeks.After 15 days of administration,fecal samples were collected from each group,and metabolomics techniques based on liquid chromatography mass spectrometry were used for the study.Results:PCA and PLS-DA scoring maps showed that there were significant differences in metabolic patterns among the three groups.Compared with the model group,the levels of Neuraminic acid,Allolithocholic acid,Alanyltryptophan,3,9-hexadecadiynoic acid,N-Palmitoyl tyrosine,Linoelaidyl carnitine,3-Hydroxy-cis-5-tetradecenoylcarnitine in Shenmai injection group were reversed.Conclusion:The mechanism of Shenmai injection intervention in hypertensive heart failure involves restoring amino acid metabolism,protein metabolism,carnitine metabolism,bile acid metabolism,lipid metabolism and etc.
