原花青素对TFA染毒小鼠睾丸组织及其激素损伤的修复作用

摘    要:目的:探讨原花青素(PC)对反式脂肪酸(TFA)染毒小鼠睾丸组织及其激素损伤的修复作用。方法:50只昆明种雄性小鼠随机等分为5组:溶剂对照组、TFA染毒组、TFA染毒+低、中、高剂量PC保护组。TFA染毒组小鼠按100.0 mg/(kg·bw)剂量灌胃TFA;低、中、高剂量PC保护组小鼠分别按25、50、100 mg/(kg·bw)剂量灌胃PC溶液。对照组灌胃生理盐水,其它各组均上午灌胃TFA、保护组下午灌胃PC溶液,每天1次,实验时间均为90 d。实验毕处死动物取血及双侧睾丸测定相关指标。结果:TFA染毒组睾丸组织形态结构损伤严重,出现基底膜模糊不清、核间隙增宽,支持细胞水肿、破裂,精子数少等病变;给予不同剂量PC保护后睾丸的结构受损较TFA染毒组有明显所改善,以高剂量PC改善明显;TFA染毒组小鼠血清FSH、LH、T值及睾丸中SOD活性均低于对照组,而MDA含量高于对照组(P0.05);经不同剂量PC保护后小鼠血清FSH、LH、T值及睾丸中SOD活性均高于对照组,而MDA含量低于对照组(P0.05)。结论:过量摄入TFA可致小鼠睾丸及雄激素受损;经一定剂量的PC保护后其受损的睾丸结构及雄激素有修复作用,其作用机制可能与抗氧化损伤有关。

Repair effect of procyanidins on Testicular tissue and androgens Injury by Tans Fatty Acids in male mice

Abstract:Objective:To explore the repair effect of procyanidins on Testicular tissue and androgens Injury by Tans Fatty Acids in male mice. Methods:50 adult Kunming (KM) mice were randomly divided into 5 groups: solvent control group, TFA contaminationgroup, TFA and low (medium, high) dose PC groups. TFA contamination group intragastric administration TFA is at the dose of 100 mg/( kg· bw). TFA and low, mid, high doses PC groups intragastric administration TFA is at the dose of 100 mg/(kg·bw), and PC at the doses of 25, 50, 100 mg/(kg· bw), respectively; Solvent control group intragastric administration is at the equal volume of physiological saline; Other groups intragastric administration TFA is in every morning, protection group intragastric administration PC is in every afternoon, last 90 days. At end of the test,obtained the blood and Bilateral testes to determinate the related indicators. Resuh:Testicular tissue morphological structure of TFA contamination group were Badly damaged. There were the Basement membrane slur, The nuclear gap widening, cellular edema, cell rupture, Less sperm count. After given different doses of PC protection, there was a significant improvement in The damage of Testicular tissue morphological , specially TFA + high - dose PC group improved obviously. Compared with solvent control group , the FSH,LH,T value of TFA contamination in the serum and SOD acitivity in the testis was decreased, while MDA content was increased ( P 〈 0.05 )., but after the Different dose of TFA + PC groups, FSH, LH,T value and SOD acitivity was increased, while and MDA content was decreased (P〈 0.05 ). Conclusion:The Excessive intaking TFA would injure Mice testicles and androgen, After a dose of PC protection, Testicular structure and androgen can be repaired, Its mechanism of action may be associated with resistance to oxidative damage.
