ADMA对人单核/巨噬细胞MCP-1及LOX-1表达的影响

摘    要:目的通过观察不对称二甲基精氨酸(ADMA)对单核细胞转化为巨噬细胞过程中分泌单核细胞趋化蛋白-1及植物凝集素样氧化低密度脂蛋白受体-1的影响,探讨ADMA在单核细胞转化为巨噬细胞过程中的作用。方法分别用ADMA、ADMA及L-精氨酸孵育单核细胞48h,酶联免疫吸附法测定上清液中单核细胞趋化蛋白-1的含量,Western blotting法测定细胞中植物凝集素样氧化低密度脂蛋白受体-1的蛋白表达量,逆转录聚合酶链反应法测定单核细胞趋化蛋白-1及植物凝集素样氧化低密度脂蛋白受体-1mRNA的表达。结果 ADMA能够促进单核细胞中单核细胞趋化蛋白-1的分泌(p〈0.01),以及上调单核/巨噬细胞中植物凝集素样氧化低密度脂蛋白受体-1蛋白的表达(p〈0.05),增加单核/巨噬细胞中单核细胞趋化蛋白-1mRNA(p〈0.01)及植物凝集素样氧化低密度脂蛋白受体-1mRNA的表达(p〈0.05),而L-精氨酸能抑制ADMA的作用(p〈0.05)。结论 ADMA能够增加单核细胞向巨噬细胞转化过程中单核细胞趋化蛋白-1及植物凝集素样氧化低密度脂蛋白受体-1的表达,并且这种作用能被L-精氨酸所抑制。

Effect of ADMA on the Expression of LOX-1 and MCP-1 in monocyte/macrophage

Abstract:Objective we aimed to explore influence of ADMA regulation on monocytes/macrophages chemotaxis by observing the effect of Asymmetric dimethylarginine (ADMA) on the expression of monocyte chemotactic protein-1 (MCP-1) and lectin-like oxidized low density lipoprotein receptor-1 (LOX-1). Methods When ADMA or ADMA plus L-arginine (L-Arg) were incubated with monocytes for 48 hs, the levels of MCP-1 in concentration medium were measured by ELISA, the protein expression of LOX-1 in cells were measured by western blot, the mRNA expression of MCP-1 and LOX-1 in cell measured by RT-PCR. Results ADMA elevated the level of MCP-1 (p0.01) and the protein expression of LOX-1 (p0.05), increased the mRNA expression of MCP-1 (p0.01) and the mRNA expression of LOX-1 (p0.05), but these effects were dismissed after giving the L-Arg (p0.05).Conclusion ADMA may increase monocytes/macrophages chemotaxis, and L-Arg can antagonize the effects of ADMA.
