CTLA—4Ig治疗C57BL／6小鼠自身免疫性肝炎的实验研究

目的 研究CTLA-4Ig在治疗C57BL/6小鼠自身免疫性肝炎上的作用。方法 通过用C57BL/6近交系小鼠的肝特异性抗原与弗氏完全佐剂混合物免疫攻击小鼠，随后用CTLA-4Ig治疗，观察小鼠的临床经过、血生化、肝脏组织学改变。结果 随着免疫次数的增多，治疗组临床经过、血生化、肝脏组织学改变逐步与正常对照相似，而病理模型组与前两组有明显差异；血生化可见天冬氨酸氨基转移酶、球蛋白显著升高；肝组织学检测可见炎细胞的浸润，肝细胞的肿胀、灶性坏死甚至广泛坏死；肝组织免疫荧光检测提示有大量免疫球蛋白沉着。结论 CTLA-4Ig能有效地治疗C57BL/6小鼠自身免疫性肝炎。
