喉癌相关基因 LCRG1蛋白的细胞亚定位及生物学特性研究

摘    要:目的:研究我们实验室最近克隆的喉癌相关基因LCRG1编码蛋白的细胞定位以及其是否具有抑瘤功能。方法:采用绿色荧光蛋白(GFP)荧光定位的方法,研究该基因编码蛋白的细胞亚定位;通过绘制细胞生长曲线,以及运用转染细胞软琼脂集落形成试验和裸鼠致瘤试验,研究喉癌相关基因LCRG1在喉癌细胞系Hep-2中的生物学特性。结果:喉癌相关基因LCRG1编码的蛋白定位在细胞核内;将CLRG1cDNA导入到不表达该基因的喉癌细胞系Hep-2中,观察到明显的生长抑制作用,表现为抑制Hep-2细胞的停泊非依赖性生长和抑制裸鼠致瘤性。结论 :喉癌相关基因LCRG1具有一定的抑瘤功能。

Abstract:Objective:To investigate the subcellar location of the protein encoded by LCRG1(Laryngeal Carcinoma Related Gene 1) and whether it has tumor suppressor function. Methods: The subcellar location of the protein encoded by LCRG1 was detected by constructing the plasmid to express the EGFP C1 LCRG1 fusion protein; the biological characteristics were studied by cell transfection,cell growth curve,colonies formation analysis,and tumorigenicity experiments. Results: The protein encoded by laryngeal carcinoma related gene 1(LCRG1) is localized in nucleus. We also transfected the wild type gene into LCRG1 negative laryngeal carcinoma cell line (Hep 2) and it potently inhibited tumorigenesis in vitro and in vivo,as shown by the dramatic growth arrest observed in a colony assay and the suppression of anchorage independent growth potential and tumor formation in nude mice. Conclusion: These data suggest that LCRG1 may be a candidate of tumor suppressor gene.
