FHIT、MLH1及Ki67与宫颈癌的关系

摘    要:目的:探讨FHIT、MLH1及Ki67在宫颈鳞癌发生、发展中的作用和意义,以及三者之间的关系。方法:应用免疫组化方法检测50例宫颈癌和20例正常宫颈组织中FHIT,Ki67和MLH1蛋白的表达。结果:MLH1蛋白的表达随病理分级的升高而降低(P〈0.05),与年龄、临床分期、组织学类型、淋巴结转移无明显相关性。FHIT蛋白的表达与MLH1的表达成正相关(rs=0.340,P〉0.05),与Ki67的表达成负相关(rs=-0.292,P〈0.05)。结论:FHIL、MLH1及Ki67在宫颈鳞癌中的表达可作为预后指标;三者有协同作用,共同检测有助于预测宫颈癌的侵袭、转移、预后。

Abstract:Objective:To investigate the expression of FHIT,MLH1 and Ki67and their relationship with clinicopathologic parameters and to investigate the relationship among them.Methods:Immunohistochemical analysis of FHIT,MLH1 and Ki67 was performed in 47 cases of invasive cervix carcinoma,26 of CIN and 20 of normal cervix tissue.Results:Compared with normal cervical tissue,the expression of FHIT protein was significantly reduced or absent in invasive cervical carcinoma((P<0.01)).Reduction or absence of FHIT expression was found in 57.45%(27/47)of the tumors.Reduction or absence of MLH1 expression was found in 65.99%(31/47)of the tumors.Positive staining of Ki67 was detected in 34 of 47(72%) cervical carcinoma.Loss of FHIT protein was closely related to pathologic grade((P<0.05)) and lymph node metastasis(P<0.01),histologic type(P<0.01),but not to stage((P>0.05)).Loss of MLH1 protein was closely related to stage,but not to pathologic grade,lymph node metastasis and histologic type(P>0.05).The expression of Ki67 in the cervix carcinoma was closely related to stage,but not to pathologic grade,lymph node metastasis and histologic type(P>0.05).There was a positive correlation between FHIT and MLH1(rs=0.340,P<0.05),and a negative correlation between FHIT and Ki67(rs=-0.292,P<0.05).Conclusion:The expression of FHIT,MLH1 and Ki67 in normal cervical epithelia,CIN,and the cervix carcinoma is gradually increased.And FHIT,MLH1 and Ki67 can play important role in the development of cervical carcinoma.Simultaneous detection of FHIT,MLH1 and Ki67 conduces to the prediction of the invasion,aversion and prognosis of cervical carcinoma.
