Expressions of miR-181a-5p and miR-126 in the serum of patients with inflammatory bowel disease and their correlation with intestinal flora

Abstract:Objective This paper intends to explore the expressions of miR-181a-5p and miR-126 in the serum of patients with inflammatory bowel disease(IBD)and their correlation with intestinal flora.Methods A total of 100 patients with active IBD admitted to Baoding Second Hospital from October 2016 to December 2018 were assigned to the ulcerative enteritis(UC)group(n=60)and the Crohn′s disease(CD)group(n=40)based on the types of IBD,while another 50 healthy volunteers who had a physical examination in the hospital during the same period were assigned to the control group.Real-time qPCR was used to detect the expressions of serum miR-181a-5p and miR-126,and patients with IBD were divided into miR-181a-5p high expression groups(n=70)and miR-181a-5p low expression group(n=30),miR-126 high expression group(n=80)and miR-126 high low expression group(n=20).The intestinal flora of each group was cultured and analyzed.The Pearson method was used to analyze the correlation between the expressions of miR-181a-5p and miR-126 in the serum and the number of flora in IBD patients.Results Compared with the control group,the expressions of miR-181a-5p and miR-126 in the serum of the study group,the UC group,and the CD group were significantly increased(all P<0.05).Compared with the control group,the numbers of Enterococcus,Saccharomyces,Bacteroides,Bifidobacterium,Peptococcus,Lactobacillus,and Clostridium were significantly increased in the UC group(P<0.05).Besides,the numbers of genus Bacteroides,Bifidobacterium,Peptococcus,and Lactobacillus increased significantly(P<0.05).Compared with the control group,the culture-positive rates of Bacteroides,Bifidobacterium,Peptococcus,and Lactobacillus in the UC group were significantly increased(P<0.05).Compared with the control group,the culture-positive rates of Saccharomyces,Bifidobacteria,Peptococcus,and Lactobacillus in the CD group were significantly increased(P<0.05).Compared with the miR-181a-5p low expression group,the numbers of Enterococcus,Saccharomyces,Staphylococcus in the miR-181a-5p high expression group were significantly increased(P<0.05).Compared with the miR-126 low expression group,the numbers of Enterococcus,Bifidobacteria,Peptococcus,and Staphylococcus in the miR-126 high expression group were significantly increased(P<0.05).The expression of miR-181a-5p was negatively correlated with Enterococcus,Saccharomyces,Bifidobacterium,and Enterobacter(P<0.05),while the expression of miR-126 was negatively correlated with Peptococcus and Staphylococcus(P<0.05).Conclusion The expressions of miR-181a-5p and miR-126 in the serum of IBD patients are increased,both of which are closely related to the changes in the composition of Enterococcus and Saccharomyces in the intestinal tract of IBD patients.
