己基可可碱对成纤维细胞的生物学影响

摘    要:目的 探讨己基可可碱对成纤维细胞的生物学作用及其意义。方法 通过MTS/PMS法、流式细胞术、3H-胸腺嘧啶摄入法、3H -脯氨酸摄入法的实验 ,观察了己基可可碱对体外培养的增生性瘢痕成纤维细胞的增殖活性和胶原合成的作用。结果 己基可可碱能够抑制瘢痕成纤维细胞的增殖和胶原的合成 ,并且具有明显的时间 -剂量依赖性 ,随着作用时间的延长和作用剂量的增大 ,抑制率逐渐增高。结论 己基可可碱具有抑制瘢痕成纤维细胞增殖和胶原合成的作用 ,从而达到抑制瘢痕增生的作用

Abstract:Objective To observe the effects of pentifylline on fibroblasts derived from human hypertrophic scar. Methods Fibroblasts derived from human hypertrophic scar were cultured in vitro. The effects of pentifylline on cell proliferation and collagen synthesis of fibroblasts were evaluated by MTS/PMS method, flow cytometry and 3 H thymidine uptake assay and 3 H proline uptake assay. Results It demonstrated that pentifylline can inhibit cell proliferation and collagen synthesis of fibroblasts derived from pathologic scar in a time-dose dependent manner. Such effects were increased while the concentration of the drugs was raised or the given time of action was prolonged. Conclusion Pentifylline can significantly inhibits the proliferation and collagen synthesis of hypertrophic scar fibroblast in vitro, which may have some effect in ameliorate pathologic scar formation.
