尼莫通对脑缺血再灌注细胞凋亡相关基因表达影响

摘    要:①目的探讨尼莫通对脑缺血再灌注时细胞凋亡的影响。②方法对实验大鼠于脑缺血前应用尼莫通进行干预,然后采用免疫组化法检测脑缺血再灌注不同时间内凋亡相关基因c-los及BCL-2蛋白表达的变化。③结果尼莫通组大鼠脑缺血后的梗死体积显著减小,脑缺血再灌注时皮质和基底核区c-los的表达明显下降,而BCL-2的表达明显增高(t=2.482,x^2=229.939~719.966,P〈0.05)。④结论尼莫通可抑制脑缺血再灌注时细胞凋亡的发生,具有神经保护作用。

Abstract:Objective To investigate the effect of Nimodipine on expression of related gene in apoptosis after focal cerebral ischemia followed by reperfusion. Methods Rats were pretreated with Nimodipine prior to MCAO, changes of protein expression of c-fos and BCL-2 in rats after ischemia and reperfusion were observed by immunohistochemical method. Results The infarct volume in Nimodipine group was significantly smaller than that in the corresponding regions in the control group. In Nimodipine group, the expression of c-fos decreased, but BCL-2 increased significantly after ischemia/reperfusion both in the ischemic cortex and in the ischemic basalganglia(t=2.482,~2=229.939-719.966,P<0.05). Conclusion Nimodipine might inhibit the development of apoptosis inrats after ischemia and reperfusion and have the neuroprotective effect against ischemia
