燕麦β-葡聚糖对高脂血症大鼠肝损伤的保护作用

目的研究燕麦β-葡聚糖对高脂血症大鼠肝损伤的保护作用。方法健康成年SD大鼠70只,雌雄各半,随机分成正常对照组(n=10)和模型组(n=60)。模型组喂高脂饲料,3w后通过对实验大鼠进行血脂检测、B超检查和随机挑选性解剖,确定模型组的肝损伤。挑选50只肝损伤大鼠,按血脂随机分为5组(n=10):燕麦β-葡聚糖低、中、高剂量组133、266和533 mg/(kg bw.d),灌胃],高脂对照组(灌胃等量蒸馏水),降脂对照组壳寡糖200 mg/(kg.d),灌胃]。正常对照组喂普通饲料、蒸馏水4 w,其它各组继续喂高脂饲料4 w。分别于大鼠灌胃给药前及给药4 w眼眶采血,测定血清ALT、AST和MDA;末次采血后取大鼠肝脏标本并检测其肝脏MDA指标。结果与高脂对照组相比,燕麦β-葡聚糖高剂量组的血清ALT、AST明显降低,血清及肝脏MDA水平下降最明显,中剂量组和壳寡糖组MDA下降水平次之;低剂量组下降无统计学意义。三个剂量组肝细胞变性、坏死明显轻于对照组。结论燕麦β-葡聚糖对大鼠高脂血症所造成的肝损伤有保护作用。

THE HEPATOPROTECTIVE EFFECT OF OAT BETA-GLUCANS ON LIVER INJURY IN HYPERLIPEMIA RATS

Objective To investigate the therapeutic effect of oat β-glucans on liver injury in hyperlipemia rats.Method Rats were fed on high fat forage for 3 w to establish hepatic injury model,then the model group was orally given oat β-glucans at doses of 133,266 and 533 mg/(kg bw.d) respectively.At the same time,there were 3 control groups including 200 mg/(kg bw.d) chitosan oligosaccharide,normal and liver injury(model group fed with distilled water).Serum levels of ALT and AST,the content of MDA in serum and liver homogenate was assayed.Results Oat β-glucans at high dose obviously decreased the levels of ALT,AST and MDA in the liver injury models,SP had a better protective effect on the liver injury than oligosaccharide did.Besides,in the treated group with β-glucans,degenerative and necrotic changes of the liver cells were apparently milder than those of the untreated group.Conclusion Oat β-glucans can obviously relieve the liver injury in hyperlipemia rats.
