紫外线对翼状胬肉细胞的作用

摘    要:目的研究紫外线对翼状胬肉细胞的作用,探讨紫外线和翼状胬肉发生发展的关系。方法收集翼状胬肉标本,采用血管内皮细胞和成纤维细胞单独培养、条件培养和共同培养的方法构建体系,用MTY法选择紫外线照射细胞的适宜强度;采用M1Tr法绘制强度20mJ/cm2紫外线照射下细胞生长曲线;采用ELISA和RT—PCR检测紫外线照射下3种体系中细胞上清液和细胞中血管内皮细胞生长因子(VEGF)和成纤维细胞生长因子(bFGF)的蛋白和RNA含量变化。结果不同强度紫外线照射细胞活性实验,选择20mJ/cm2强度作为实验照射基数;翼状胬肉微血管内皮细胞(PE)和成纤维细胞(PF)在紫外线照射下,细胞活性下降;单独培养PE受紫外线照射后VEGF的RNA含量和上清液VEGF蛋白含量,照射组较未照射组有显著性下降(P〈0.05);单独培养和条件培养PF受紫外线照射后VEGF的RNA含量和上清液VEGF含量,照射组较未照射组有显著性下降(P〈0.05)。3种体系PE和PF照射后bFGF的RNA含量及细胞上清液中bFGF的蛋白含量,照射组较未照射组呈显著性下降(P〈0.05)。结论紫外线对体外培养的血管内皮细胞和成纤维细胞生长有抑制作用。

Abstract:Objective To detect the relationship between ultraviolet and pathogenesis of pterygium through study at effect of irradiated on pterygial cell models. Methods Pterygial samples were extracted and collected and the pterygial endothelial cells (PE) and pterygial fibroblasts (PF) were cultured alone, condi- tional and co-cultured to form different culture systems. The methods included that to select the suitable intensity of uhraviolet(UV) by MTT, to detect the changes of curves of growth about two kinds of cells by MTT and to explore the developments of protein and RNA of vascular endothelial growth factor (VEGF) and fibroblast growth factor-basic(bFGF) in three culture systems under ultraviolet whose intensity is 20 mJ/cm2 by ELISA and RT-PCR. Results 1. The suitable intensity of ultraviolet selected was 20 mJ/cm2. 2. We found that there were many changes of proliferation of PE and PF, included that the activities of cells are declined, speeds of growth are decreased and growth cycles are blocked. 3. We found that the protein of VEGF in supernatant and RNA of VEGF in PEC cultured alone system after irradiated were much lower than un-irradiated. The protein of VEGF in supernatant and RNA of VEGF in PF cultured alone and conditional system after irradiated were much lower than un-irradiated. 4. The protein of bFGF in supernatant and RNA of bFGF in PE and PF cultured alone, conditional and co-cultured systems after irradiated were much lower than un-irradiated. Conclusion The ultraviolet has inhibited the growth of pterygialPE and PF cultured in vitro.
