茶多酚对乙醇所致原代肝细胞损伤的保护作用研究

摘    要:目的研究乙醇对原代肝细胞的氧化损伤作用,同时探讨茶多酚对乙醇所致的原代肝细胞损伤模型的保护作用。方法分离小鼠原代肝细胞,直接以不同浓度乙醇作用于肝细胞,或以茶多酚预处理后再用乙醇作用于肝细胞,测定各组细胞存活率、抗氧化酶及丙氨酸氨基转移酶(ALT)活性的变化。结果随着乙醇浓度的增加,小鼠原代肝细胞存活率逐渐下降,抗氧化酶活性降低,转氨酶升高;而茶多酚的预处理对于乙醇所致的小鼠原代肝细胞损伤具有明显的保护作用,表现为相同乙醇浓度作用下,随茶多酚浓度的升高,细胞存活率逐渐升高,抗氧化酶活性增加,转氨酶下降,且差异有统计学意义(P<0.05)。结论乙醇可以诱导小鼠原代肝细胞的氧化损伤,而茶多酚对其氧化损伤具有明显的保护作用。

Abstract:Objective To investigate the protection of tea polyphenol (TP) against the ethanol induced damage on the mice primary hepatocytes. Methods The effect of ethanol on the cytotoxicity and activities of antioxidative enzymes as well as the activity of alanine aminotransferase (ALT) in the culture of mice primary hepatocytes. Different doses of TP were given before ethanol exposure. Results The cell viability and the activities of antioxidative enzymes were reduced while the activity of ALT enhanced with increasing of ethanol doses. Furthermore,primary hepatocytes pretreated with TP showed obvious protective effect on the cell damage induced by ethanol. At the same ethanol dose the cell viability and the activities of antioxidative enzymes were elevated while the activity of ALT decreased with increase of TP dose. Conclusion High dose ethanol can induce an oxidative damage on mice primary hepatocytes and TP pretreatment display a remarkable protective effect to adverse damage caused by ethanol.
