清开灵有效组分对缺血脑组织神经营养因子含量的影响

摘    要:目的观察清开灵有效组分对缺血不同时段脑组织神经生长因子(NGF)、碱性成纤维细胞生长因子(bFGF)、脑源性神经营养因子(BDNF)含量的影响,以期从神经营养因子环节探讨清开灵有效组分抗脑缺血损伤的机制.方法参照Longa法复制脑缺血模型,清开灵有效组分干预,酶联免疫吸附实验(ELISA)法测定脑组织NGF、bFGF、BDNF含量.结果脑缺血12 h和24 h,模型组脑组织神经营养因子含量反应性增加.脑缺血12 h和24 h栀子苷组、珍珠母水解液组脑组织NGF含量显著降低,脑缺血12 h胆酸组脑组织NGF含量显著升高;脑缺血24 h胆酸组脑组织bFGF含量显著降低,其余各用药组脑组织bFGF含量均增加;脑缺血12 h和24 h,各用药组脑组织BDNF含量均降低.结论清开灵有效组分对脑缺血损伤神经元的保护作用中,促进星形胶质细胞等产生bFGF是其主要途径.

Abstract:Objective To observe the effects of the effective fractions of Qingkailing (EFQs) on the levels of NGF, bFGF and BDNF at different intervals in the rat brain undergoing ischemia, in order to find the mechanism of EFQs against cerebral ischemic injury on the basis of neurotrophic factors. Methods The Longa's procedure was used to induce cerebral ischemia in the rat, and EFQs were given to the model for intervention, and ELISA was used to detect the levels of NGF, bFGF and BDNF in the brain of the model rats. Results The levels of the mentioned neurotrophic factors were reactively increased at the 12 th hour and 24 th hour, respectively during the cerebral ischemia in the model group; the level of NGF was markedly decreased at the 12 th hour and 24 th hour, respectively in both the geniposide group and the Concha Margaritifera Usta group; the level of NGF was markedly increased at the 12 th hour in the cholic acid group; the level of bFGF was markedly decreased at the 24 th hour again in the cholic acid group while it was increased in all other groups; and the level of BDNF was decreased at the 12 th hour and 24 th hour in all the EFQs-treating groups.Conclusion Promotion of the production of bFGF by the astrocytes is the main mechanism of EFQs in protecting neurons from being injured by cerebral ischemia.
