摘    要:目的探讨三七总皂苷(PNS)对肝纤维化大鼠肝脏超微结构的影响及意义.方法通过电镜观察大鼠肝脏的超微结构,以不同剂量PNS分别对肝纤维化大鼠进行预防和治疗.结果肝纤维化大鼠可出现肝细胞胞浆内肿胀空泡和脂滴、微绒毛稀少、窦内皮细胞肥胖、细胞器增多、Disse间隙内胶原纤维增生等超微结构改变.PNS可减轻大鼠肝纤维化模型肝细胞内脂滴形成、内质网增生、线粒体肿胀及间质胶原增生.结论无论治疗和预防组,PNS均可有效减轻肝纤维化大鼠肝脏超微结构的损伤.

Abstract:Objective To investigate the effect and significance of panax notoginoside (PNS) on the hepatic ultrastructure in hepatic fibrosis rats. Methods Hepatic fibrosis (HF) rats were established by subcutaneous injection at one hind leg of pure CCl_4 at dose of 0.5 ml/100 g for the first time and then olive oil containing 40% CCl_4 at dose of 0.3 ml/100 g twice a week for 6 weeks. The normal controls were injected with physiologic saline. HF rats were simultaneously treated by different dose of PNS as the hepatic fibrosis was establishing or treated after establishment. The hepatic ultrastructure of all rats was observed by electron microscope. Results The change of hepatic ultrastructure could be observed in HF rats. There were swelling vacuoles and fatty drops in hepatic plasma; microvilli became exiguous. The number of organelle increased in endothelial cells of hepatic sinusoid and collagen fibre proliferated in hepatic Disse space. PNS would reduce formation of lipid droplet, proliferation of endoplasmic reticulum, swelling of mitochondrion and proliferation of interstitial collagen in hepatic cells of HF rats. Conclusion PNS could relieve the damage of hepatic ultrastructure in HF rats either for the purpose of prevention or cure.
