胰岛素对血管内皮细胞衰老及Akt表达的研究

摘    要:目的:探讨胰岛素对人脐静脉内皮细胞(human umbilical vein endothelial cell,HUVEC)衰老及Akt表达的影响。方法:不同浓度的胰岛素(对照组、1 nmol/L、10 nmol/L、胰岛素100 nmol/L)加入体外培养的人脐静脉内皮细胞,24 h后β半乳糖酶染色检测细胞衰老率和免疫组化检测Akt活性。结果:与对照组比较,1 nmol/L组和10 nmol/L组细胞衰老程度降低,Akt表达上调。但随着浓度增加,100 nmol/L组细胞衰老反而加重。而Akt表达无统计学差异。结论:低浓度胰岛素可延缓内皮细胞衰老,可能与Akt基因表达上调有关。而高浓度胰岛素加重细胞衰老,其机制可能与Akt蛋白无关。

Abstract:Objective:To investigate the effects of insulin on senescence of human umbilical vein endothelial eell (HUVEC) and Akt expression. Methods:The HUVEC were incubated with different concentrations of insulin (control group, 1 nmol/L group, 10 nmol/L group,100nmol/L group). 24 hours later,the cell senescence rate was measured by SA-β-gal staining, and the expression of Akt was measured by immunohistochemistry. Results: Compared to the control group, the cellular senescence rate of 1 nmol/L and 10 nmol/L group decreased,while Akt expression increased. But with the increasing of insulin concentrations, the cellular senescence rate of 100 nmol/L insulin group increased obviously,and Akt expression unchanged. Conclusion:Low concentration insulin might inhibit human umbilical vein endothelial cells senescence ,which is related with the increase of Akt expression. High dose of insulin might enhance cel-lular senescence,which might be unrelated to Akt protein.
