半定量RT-PCR法检测TLR4在人大肠癌中的表达

目的研究TLR4 mRNA在人大肠癌组织和正常肠道组织之间的差异性表达,探讨TLR4同肠道肿瘤之间的关联性。方法以GAPDH为内参,运用半定量RT-PCR技术检测24例正常大肠组织标本和24例新鲜大肠癌组织标本中TLR4、CD14、MD2、TGF-β和VEGF因子表达水平,应用多媒体图象分析软件进行分析和比较。结果TLR4、MD2、CD14、TGF-β和VEGF在肿瘤组织中的表达量均比在正常组织中的表达量增强,并且TLR4与TGF-β、VEGF之间表达存在较强的关联性（r〉0.838,P〈0.01）。结论人大肠组织中TLR4表达量与肿瘤的发生具有一定的相关性,推测可能诱导肿瘤相关细胞因子和趋化因子的产生。

A study on the expression of TLR4 in humen colorental cancer by RT-PCR

Objective Tovalidate theexpressiondifferenceof TLR4 in normaland carcinoma oflargeintestinal tissue,explore the connection between TLR4 and Colorectal cancer development.Methods The expression of TLR4、CD14、MD2、 TGF-β、VEGF were detected by RT-PCR with GAPDH as references in fresh normal colorectal tissue in 24 cases and fresh samples of colorectal cancer from the same cases.the image analysis of results with imageProplus analysis software was used to semiquantitatively determine the differents expressions between normal and carcinoma tissues.Results The expressions ofTLR4, CD,4, MD2, TGF-β, VEGF were significantly enhanced in tumor tissue than in normal tissue. There was correlation between TLR4 and TGF-β,VEGF（r〉0.838,P〈0.01）.Conclusion The study suggested that the expression of TLR4 in human colorectal cancer concerned with the occurrenced and development of minor for it may induce the production of cytokines and chemotatic factor.
