结直肠癌肝转移动物模型的建立

摘    要:目的比较几种结直肠癌肝转移动物模型的建立方法,确定一种比较适合的结直肠癌肝转移动物模型。方法以BALB/c鼠为研究对象,随机分组,分别经脾脏、直肠、腹腔按不同剂量(0.1mL、0.2mL、03mL、1.0mL)(浓度为1×10^6个/mL)注入小鼠结肠腺癌细胞(CT26)悬液,其中腹腔组无1.0mL剂量,卡方检验比较三组动物模型组内及组间肝转移率。结果三种方法均能复制出结直肠癌肝转移动物模型,脾脏组0.1mL、0.2mL、03mL、1.0mL肝转移率分别为50.0%、77.2%、50.0%、27.0%;直肠注射组0.1mL、02mL、03mL、1.0mL肝转移率为50.0%、53.1%、16.7%、6.7%;腹腔注射组0.1mL、02mL、03mL、肝转移率为10.0%、22.2%、10.0%。结论经脾脏注射0.2mL(1×10^6个/mL)BALB/c鼠结肠腺癌细胞株(CT26)是一种建立结直肠癌肝转移动物模型成功率较高的方法。而经肛门直肠注射0.1mL或0.2mL(1×10^6个/mL)BALB/c鼠结肠腺癌细胞株(CT26)是一种建立结直肠癌肝转移动物模型简便及符合结直肠癌肝转移规律的方法。

Establishment of animal models in colorectal cancer with liver metastasis

Abstract:Objective TO define a suitable method of establishing animal model of coloreetal cancer with liver metastasis by comparing several methods. Methods BALB/c mouses were grouped randomly into 11 groups by doses of adenoearcinoma cells and methods of injection, that is 0.1 mL, 0.2 mL, 0.3 mL, 1.0 mL(colon adenocareinoma cells in 1 × 10^6/mL) by spleen, rectum, peritoneal cavity( no 1.0 mL dose group in peritoneal group). The rate of liver metastases were compared between the three groups and within each group. Results Three methods were able to replicate an animal model of colorectal cancer with liver metastases. The liver metastasis rates were 50.0%, 77.2%, 50.0%, 27.0% respectively in0.1 mL, 0.2 mL, 0.3 mL, 1.0 mL doses by spleen; there were 50.0%, 53.1%, 16.7%, 6.7% respectively in 0.1 mL, 0.2 mL, 0.3 mL, 1.0 mL doses by rectum, and 10.0%, 22.2%, 10.0% in 0.1 mL, 0.2 mL , 0.3 mL doses through peritoneal cavity. Conclusion Inject through spleen with a dose of 0. 2 mL(1 × 10^6/mL) is a effective way to establish animal model of colorectal cancer with liver metastasis. Inject through rectum with a dose of 0.1 mL or 0.2 mL( 1 × 10^6/mL) CT26 is a simple way of establish animal model of coloreetal cancer with liver metastasis , and follow the law of liver metastases of colorectal cancer also.
