人α-IFN对脐血单个核细胞FcrR的调变作用及其机理初步研究

摘    要:本文以热凝聚Ig间接免疫荧光染色及EA花结法,研究发现小剂量人IFN-α对人脐血单个核细胞Ecr受体表达有明显的促进作用(P<0.01),抗人IFN-α抗体可消除这种促进作用。鼠干扰素则无明显的促进作用。链霉蛋白酶处理可减弱Fcr受体的表达,而IFN-α可恢复该受体的正常表达。IFN-α对Fcr受体的调变作用可被哇巴因阻断。


STUDY ON THE EFFECT ON INTERFERON-INDUCED CBMC-Fcr RECEPTOR EXPRESSION AND ITS MACHANISM
Abstract:Mononuclear cells from human cord blood (CBMC) were incubated with IFN-α and tested for the expression of FcrR assessed by determining the number of the cells binding heat-aggregated human IgG (-AHG) by staining with fluorescein conjugated goat anti-human IgG or counting the number of cells forming rosettes with IgG-sensi-tized chick erythrocytes. FcrR receptor expression was found to be increased in response to IFN -αafter 3 hr. treatment, especially in small dose of IFN-a treatment (P< 0.01).The anti-Hu IFN-antiserum can depressed the enhancement.The effect of mouse IFN on human CBMC-FcrR was much less than that of human IFN-a. It was found that lower expressive activity of the pronase-treated CBMC was elevated to the normal level with treatment of IFN-α. The effect of IFN-α on CBMC-FcrR can be blocked in the presence of ouabain, an inhibitor of cell membrane ATPase responsible for the uptake of potassium and sodium.
