摘    要:目的建立大枣环磷酸腺苷(cyclic adenosine 3′,5′-monophosphate,c-AMP)含量的测定方法。方法高效液相色谱法(high performance liquid chromatography,HPLC),采用MGC18色谱柱(4.6 mm×250 mm,5μm),流动相为0.05 mol/L KH2PO4-Methanol(90:10-10:90);梯度洗脱,流速1 mL/min,检测波长为254 nm,柱温为30℃。结果 c-AMP在15.6~500.0 ng范围内呈良好的线性关系(r=1.000),平均回收率为100.35%±1.58%,RSD为1.58%。结论本方法操作简便、灵敏度高、准确性和重现性好,可作为大枣的质量控制方法。

Abstract:Objective To establish the method of determination of cyclic adenosine 3′, 5′-monophosphate(cAMP) in fructus jujubae by high performance liquid chromatography(HPLC). Methods HPLC analysis was carried out using MGC18(4.6 mm×250 mm, 5 μm). The mobile phase was 0.05 mol/L KH2PO4-methanol(90:10-10:90), Grade-Washing, the flow rate was 1.0 mL/min, Detected wavelength was set at 254 nm, column temperature was 30 ℃. Results The linear range for c-AMP was 15.6~500 ng(r=1.000), the average recovery was 100.35%±1.58%, RSD was 1.58%. Conclusion The method is rapid and accurate with good reproducibility which is used for the quality control of c-AMP in jujubae.
