缺氧缺血性脑损伤后针康法对幼鼠学习记忆能力及海马微管相关蛋白-2表达的影响

摘    要:目的探讨针康法对缺氧缺血性脑损伤(HIBD)幼鼠学习记忆能力及海马微管相关蛋白-2(MAP-2)的影响.方法将80只Wistar幼鼠随机分为5组:假手术组、模型组、头穴丛刺组、环境刺激组和针康组,每组16只.每组再按造模后14 d、28 d分别处死分为2个亚组,每组8只.采用结扎左侧颈总动脉,吸入氧体积浓度为8%的氧氮混合气体,制作HIBD新生动物模型.采用Morris水迷宫评价幼鼠学习、记忆能力,免疫组化法检测海马MAP-2阳性细胞的表达.结果水迷宫实验结果显示:模型组逃避潜伏期时间延长,穿台次数减少.在定位航行、空间探索能力方面头穴丛刺组、环境刺激组和针康组与模型组比较,差异有统计学意义(P<0.05).在定位航行、空间探索能力方面针康组分别与头穴丛刺组、环境刺激组比较有统计学意义(P<0.05).免疫组化榆结果显示:头穴丛刺组、环境刺激组、针康组海马CA1区MAP-2的表达均多于模型组(P<0.05),海马CA1区MAP-2表达针康组较头穴从刺组、环境刺激组表达增加(P<0.05).结论针康法能提高HIBD幼鼠学习记忆能力,其分子机制可能与海马CA1区MAP-2阳性细胞的表达水平增加有关.
