摘    要:应用角蛋白PKK_1、PKK_2、PKK_3、波状蛋白、结蛋白、神经纤维蛋白与胶质纤维酸性蛋白5类7种单克隆抗体研究食管上皮细胞、食管癌、肝癌、肺癌、人胚肺转化细胞、胃癌、星形胶质细胞瘤(BT_(420),BT_(325))、肌细胞、肺细胞及神经纤维细胞等11种培养细胞的中间纤维表达。结果表明,不同细胞具有不同的中间纤维表达的特异性。据此特异性可鉴定胶质细胞、神经细胞、间质细胞、肌细胞与上皮细胞的类型及鉴别正常细胞与肿瘤细胞系的性质与来源。PKK_3还可用于腺癌细胞检测。

Abstract:Intermediate filaments(IF) are one of the major components of cytoskel-eton with a diameter of 8-12nm.They are heterogenousin their biochemical compositions.Five different subclasses of IF proteins have been identified and demonstrated immunocytochemically by relevant monoclonal antibodies. We studied different expression of IFs in cultured cell lines by indirect immuno-fluorescence using monoclonal antibodies directed to cytokeratinCPKK1.PKK2. PKKs) ,vimentin,desmin,glial fibrillary acid protein (GFAP) and neurofilaments (Labsystems OY,Finland). Human embryo esophageal epitnelial cells, esophageal cancer cells(ECa!09), liver cancer cells(BEL-7402), lung squa-mous cancer cells(LTEP-78) and human embryo lung diploid cells(2BS) were positive for PKK1 and PKK2.Stomach adenocarcinoma cells(SC823) were,on the other hand, positive for PKK1 and PKK3. Human embryo muscle cells was slightly positive for desmin; neurofilaments and GFAP were demonstrated in cerebral neuron cells, and human astroglioma cells(BT420, BT325)respec-tively. The tissue type specificity of the IF expression offers a reliable way to ascertain histogenesis of cancer.
