DNA损伤修复相关因子在宫颈癌组织中的表达研究

摘    要:目的探究DNA损伤修复相关因子[毛细血管扩张性共济失调突变因子(pATM)、磷酸化组蛋白H2AX(γH2AX)]在宫颈癌组织中的表达及其诊断意义。方法选取2018年2月至2019年8月于我院就诊的50例宫颈癌患者设置为试验组,另选择同期我院体检健康者50例作为对照组。两组研究对象均进行免疫组化染色,检验pATM、γH2AX的表达情况;通过磷酸化抗体检测手段检测两组研究对象的pATM、γH2AX水平。结果试验组的pATM、γH2AX mRNA的相对表达量明显高于对照组(P<0.05)。试验组pATM、γH2AX的阳性表达率明显高于对照组(P<0.05)。pATM、γH2AX高表达与宫颈癌的临床分期相关(P<0.05),与年龄、病理分化程度、化疗周期无关(P>0.05)。结论DNA损伤修复相关因子pATM、γH2AX在宫颈癌中高表达有利于疾病的的早期诊断和临床分期诊断,具有一定的临床价值。

Abstract:Objective To study the expression of DNA damage repair related factors[phosphorylated ataxiatelangiectasia mutation factor(pATM),phosphorylated histone H2AX(γH2AX)]in cervical cancer tissues and its diagnostic significance.Methods Fifty cases of cervical cancer treated in our hospital from February 2018 to August 2019 were selected as experimental group,50 healthy people who underwent physical examination in our hospital during the same period were selected as control group.The expression of pATM andγH2AX of objects in the two groups were examined by immunohistochemistry;the levels of pATM andγH2AX of objects in the two groups were detected by means of phosphorylated antibody detection.Results The relative expression of pATM andγH2AX mRNA in the experimental group were significantly higher than those in the control group(P<0.05).The positive expression rates of pATM andγH2AX in the experimental group were significantly higher than those in the control group(P<0.05).The high expression of pATM andγH2AX were correlated with the clinical stage of cervical cancer(P<0.05),but which were not correlated with age,pathological differentiation degree and chemotherapy cycle(P>0.05).Conclusion The high expression of DNA damage repair related factors pATM andγH2AX in cervical cancer are beneficial to the early diagnosis and clinical stage diagnosis of the disease,which have certain clinical values.
