摘    要:目的研究砷对原代培养大鼠海马神经元的直接毒性作用。方法在海马神经元原代培养的基础上给予不同浓度的亚砷酸钠(0,0.04,0.40,4.00,10.00 mg/L)分别处理8 h和24 h后,观察神经元活力、神经元形态学以及细胞内代谢活动的变化。结果亚砷酸钠可致体外培养的大鼠海马神经元形态结构改变、细胞活力降低、培养上清液中乳酸脱氢酶(LDH)水平升高和丙二醛(MDA)水平升高。结论亚砷酸钠对培养的海马神经元有直接毒性作用且呈剂量-时间依赖关系。

Abstract:Objective To study the direct toxicity of sodium arsenite on hippocampal neurons in rats.Methods Primarily cultured hippocampal neurons in newborn rats were exposed to different doses of sodium arsenite(0,0.04,0.40,4.00 and 10.00 mg/L)for 8 h and 24 h,and then the alterations of viability,cellular morphology and intracellular metabolic activity were examined.Results Treatment with sodium arsenite caused altered morphology,decreased cell vigor,and enhanced LDH activity and increased content of MDA in the cultured neurons as compared with those of the control.Conclusions Sodium arsenite displays direct toxicity on primary cultures of hippocampal neurons with dose-time dependent relationship.
