豚鼠肠系膜淋巴管平滑肌细胞的培养

目的 :培养豚鼠肠系膜淋巴管平滑肌细胞。方法 :肠鼠肠系膜淋巴管用胰蛋白酶消化二次 ,第一次消化 15min ,除去肠系膜、淋巴管外膜等组织 ,第二次消化中膜的平滑肌 ,3 0～ 45min后 ,吸出未消化的组织 ,获得平滑肌细胞。常规培养、传代。结果 :培养细胞经光镜、电镜和荧光显微镜可观察到平滑肌的典型的形态和结构。结论 :所培养细胞为平滑肌细胞 ,此法可应用于管径较小的淋巴管和血管平滑肌细胞的培养。

Tissue Culture of Smooth Muscle Cells of Lymphatics in Guinea Pig

Objective: To culture the smooth muscle cells of lymphatic.Method: The Guinea pig′s mesenteric lymphatic vessels was digested with trypsin for two times.Once for 15 minutes,to get rid of the mesentery and adventitia of lymphatic,go on to digest the smooth muscle cells of media with new trypsin solution for 30～45 minute.Take out the tissue left undigested,the cell suspension is centrifuged,and the cells was suspended in DEEM supplemented with 20% new calf serum(NCS),maintained at 37℃ in a humidified incubator with 95% air and 5% CO_2.Results: By the light,fluorescent and transelectron microscopy,the characteristic feature of smooth muscle cell can be seen.Conclusion: The cells in culture is smooth muscle cell.The method can be used in tissue culture of smooth muscle in lymphatic and blood vessel with minor diameter.
