摘    要:目的探讨RNAi(RNA interference,RNAi)对人胃癌BGC-823细胞中肝素酶(heparanase,HPA)mRNA表达的影响。方法针对HPA不同基因位点设计合成寡核苷酸然后转录为小干扰RNA(siRNA),分别用浓度为10μmol/L、15μmol/L、20μmol/L的siRNA经LipofectamineTM2 000脂质体介导转染BGC-823细胞72h,同时设无关序列组及不转染组。采用完整细胞原位杂交技术观察各组BGC-823细胞中HPA mRNA的表达。结果 siRNA可有效抑制人胃癌BGC-823细胞中HPA mRNA的表达,以20μmol/L的siRNA作用效应最强,但不同浓度siRNA两两相比,差异均无统计学意义(P〉0.05),不同浓度siRNA对HPA mRNA表达的抑制效应均显著强于无关序列组及不转染组(P〈0.05)。结论 RNAi技术可有效抑制人胃癌BGC-823细胞中HPA mRNA表达,为进一步运用RNAi技术治疗胃癌等恶性肿瘤奠定了一定的理论基础。

Abstract:Objective To investigate the effects of RNAi on mRNA expression of heparanase in human gastric carcinoma BGC-823 cells.Methods HPA siRNA against different heparanse mRNA sites were transfected into BGC-823 cells,the concentration of transfection are 10μmol/L,15μmol/L,20μmol/L,the time of the transfection was 72 hour,and the other group is not transfected.Then the expression of heparanase mRNA in BGC-823 cells was studied by in situ hybridization.Results The expressing of heparanase mRNA could be inhibited by siRNA,especially 20μmol/L.There was no significant among siRNA with different concentrations(P0.05),but the inhibitory effects to HPA was stronger than that to non-sense sequence group and non-transfected group(P0.05).Conclusion The expression of heparanse mRNA in BGC-823 cells could be inhibited by siRNA,these results provide the basic theory to prevent the invasion and metastasis of gastric carcinoma.
