P-gp、p53过表达与胃癌及乳腺癌转移的关系

摘    要:目的研究胃癌和乳腺癌组织P-耐药糖蛋白(P-gp),p53蛋白表达及临床意义.方法应用免疫组化SABC法检测93例胃腺癌组织及60例乳腺癌组织中p-gp、p53蛋白的表达.结果乳腺癌组织中P-gp阳性率为38.3%,p53阳性率为56.7%;胃腺癌组织中P-gp阳性率为55.9%,p53阳性率为44.1%;P-gp、p53蛋白表达均与淋巴结转移有关(P<0.05)P-gp表达与p53表达有显著的协同性(P<0.05).结论p53突变蛋白和P-gp过表达在胃癌和乳腺癌淋巴结转移中起重要作用.p53基因突变对于多药耐药基因(mdr-1)编码产物P-gp表达增高有一定影响,早期进行p53蛋白和P-gp检测有重要的临床意义.

Abstract:Objective To study the level of p53 and P-glycoprotein (P-gp) and its clinical significance in tissues of gastric cancer and breast cancer. Methods 93 adenocaroinomas and 60 breast cancer specimens were analyzed for p53 and P-gp by immunohistoche-mical staining. Results In breast carcinoma the positive expressions for P- gp and p53 were 38.3 and 56.7% respectively. In gastric carcinoma the positive expressions of P-gp and p53 were 55.9% and 44.1% respectively. A significant correlation was found that both P-gp and p53' s over-expression were related to lymph node metastasis (P < 0.05) . Co-expression of p53,and P-gp were commonly (P <0.05). Conclusion Mutation of p53 and P-gp over-expression may pky an important role in the lymph node metastasis of gastric cancer and breast cancer. Mutation of p53 gene may affect the over-expression of mdr-1 gene activity, so that p53 and P-gp gene products immunostaining assay at the early stage of gastric cancer and breast cancer may be of great clinical significance.
