Abstract: | The in vitro effects of a homologous series of alkyl 2-cyanoacrylate monomers on the conversion of 14C-labeled acetate to respiratory 14CO2 by rat liver cytoplasm preparations were determined. The labeled preparation was added to measured amounts of monomer in vessels equipped with center wells containing p-(diisobutul cresoxyethoxyethyl) dimethylbenzylammonium hydroxide (methyl benzethonium chloride). The mixture was incubated at 37° for 45 min; metabolism was terminated with 1 n H2SO4, and the amount of 14CO2 produced was measured. The methyl homolog caused the greatest inhibition. In general, an increase of the length of the alkyl chain caused a decrease in the inhibitory effect. These results generally corroborate findings of in vivo histotoxicity studies. While this technique has been used in the past for determining pharmacologic activity, it also offers a simple and rapid means of predicting potential toxicity. |
